"The center" is not one place, and the answer changes
Buy a ticket before you board: machines at the airport stops, the DPP info centre in each arrivals hall (daily 07:00–21:00), or the PID Lítačka app, activated at least 60 seconds before you get on. Stamp a paper ticket once, at the validator on the platform — contactless card readers on board work too.
The taxi rank
The official taxi is Uber Airport: it shows the price before you confirm and can't exceed the regulated maximum, so book it rather than take a ride offered inside the terminal. If any other driver asks for more than the price shown or won't give a receipt, don't pay — call the municipal police on 156.
Uber and Bolt
Uber, Bolt and Liftago all serve the airport, but not at the official Uber Airport spot in front of arrivals — their pickup point is the car parks P1 EXPRESS, P2 EXPRESS or PB EXPRESS, signed "Taxi Point" at P1 and PB.
Which terminal
Land at Terminal 2 and the Airport Express won't help — toward the city it leaves only from directly in front of Terminal 1. The trolleybus and the ordinary buses stop at both terminals, so use one of those rather than crossing over just for the express.

The express takes 47 minutes on weekday daytime runs (37 early/late, 34 late evening, 30 at night) — no faster than the trolleybus-and-metro's roughly 35 minutes, for four times the price.
Nothing you already hold works on it: not a 24- or 72-hour ticket, not a pass, not an SMS ticket. Children under 15 pay CZK 100 here and nothing on the ordinary network.
It runs all night, hourly: 00:37, 01:37, 02:37 and 03:37 from Terminal 1, then 04:35 and 05:35, then every 20 to 30 minutes by day. The longest wait is the 62 minutes between 23:35 and 00:37.
At the main station its stops are in Opletalova street, about 50 metres past the Jeruzalémská junction, until the Wilsonova roadworks end — expected November 2026. The airport's page still describes the old stop outside the Fanta building.
The night departures set down at Náměstí Republiky 20 minutes out, where the night trams meet; the daytime ones run non-stop.
Boarding is through the front door only; buy from the driver if you have no ticket.

It ends in the New Town, not at the main station or the Old Town: I. P. Pavlova is a ten-minute walk from Wenceslas Square. For Náměstí Republiky or Hlavní nádraží wait for the 907, or pay CZK 200 for the express.

Worth knowing
The Airport Express costs CZK 200 and takes 47 minutes to the main station. Trolleybus 59 and metro A cost CZK 50 and reach Můstek, at the foot of Wenceslas Square, in about 35 minutes — cheaper and faster.
Only the express's own CZK 200 ticket (CZK 100 for a child 6–14) works on it — no PID pass, and not the free travel Prague gives under-15s and over-65s. Two adults and two children pay CZK 100 total by trolleybus and metro, but CZK 600 by express.
The 30-minute ticket (CZK 39) will not last: the trolleybus alone takes 17 minutes, plus nine or ten more on the metro with a change. Buy the 90-minute ticket instead: CZK 50 on paper or at the machine, CZK 46 in the PID Lítačka app.
Ordinary PID lines carry bags free only up to 25 × 45 × 70 cm; anything larger needs a luggage ticket, CZK 20–25 (sources disagree — budget for CZK 25). A 24- or 72-hour ticket includes one such bag free. Luggage is free on the Airport Express.
There is no night gap. The Airport Express runs hourly all night: 00:37, 01:37, 02:37, 03:37, then 04:35. On the ordinary CZK 50 ticket, night bus 910 runs every 30 minutes, 23:51–03:54; night bus 907 hourly, 01:05–04:03. Land at 03:00 and four buses arrive within the hour.
No taxi fare can be stated: Prague sets a maximum meter rate — boarding charge, per-kilometer rate, per-minute waiting — regardless of party size, so a bigger group never makes it cheaper. Expect around CZK 800 for the roughly 20 km to the center, though this is an estimate, not a fixed fare. Book the official Uber Airport at kiosks in both arrivals halls; it shows a price before booking.
The Airport Express leaves from Terminal 1 only — Terminal 2 arrivals must walk over. At the main station, it stops on Opletalova street, not outside the Fanta building, until roadworks end, expected November 2026.
